Angelman syndrome usually results from loss of functional UBE3A activity in brain neurons, where the maternally inherited copy is normally the active copy. Several genetic mechanisms can cause that loss, including deletion of the maternal chromosome 15 region containing UBE3A, a variant in the maternal gene, paternal uniparental disomy, or an imprinting defect.
